佐保子","creatorNameLang":"ja"}],"nameIdentifiers":[{}]}]},"item_10001_description_5":{"attribute_name":"内容","attribute_value_mlt":[{"subitem_description":"近年,光線力学療法の有効性が認められ,種々の悪性腫瘍への適応が検討されてきている。しかしながら,この光線力学療法の殺細胞作用機序に関する細胞レベルでの研究は,いまだ多くの点が不明のままである。そこで我々は,本治療法で用いられる光感受性腫瘍親和性物質を培養腫瘍細胞に投与し,主として免疫蛍光抗体法と電子顕微鏡法を用いた形態学的アプローチによる,細胞死誘発作用機序の解明を試みた。光感受性腫瘍親和性物質投与後,アクロレインを用いて急速に固定された細胞の電子顕微鏡観察によって,特にミトコンドリアのクリスタ構造と粗面小胞体嚢の構造に変化がみられた。これらの構造変化は,用量依存的かつ経時的な変化であり,光照射後30分で,細胞死が誘発された。誘発されたこれらの細胞死に,アポトーシスを思わせる所見は確認できなかった。ミトコンドリアと粗面小胞体などの細胞質細胞小器官は細胞内膜構造を有しており,この内膜の膜蛋白を介して,細胞骨格であるアクチンフィラメント,中間径フィラメント,微小管と結合している。そこで,βチューブリン,非筋型アクチン,サイトケラチン,ビメンチンに特異的な抗体を用いて,微小管,アクチンフィラメト,中間径フィラメントの分布の変化を観察したところ,特にアクチンフィラメントに大きな変化が認められた。通常の1/10の濃度である光感受性腫瘍親和性物質投与後,さらにアクチンフィラメント細胞骨格毒であるサイトカラシンBを0.1μg/ml追加投与し,ハロゲン光下に置いたところ,通常では殺細胞効果がほとんど期待できないような低濃度の光感受性腫瘍親和性物質濃度でも,十分な殺腫瘍細胞効果","subitem_description_language":"ja","subitem_description_type":"Other"}]},"item_10001_identifier_registration":{"attribute_name":"ID登録","attribute_value_mlt":[{"subitem_identifier_reg_text":"10.34429/00002928","subitem_identifier_reg_type":"JaLC"}]},"item_10001_publisher_8":{"attribute_name":"出版者","attribute_value_mlt":[{"subitem_publisher":"山梨医科大学医学会","subitem_publisher_language":"ja"}]},"item_10001_source_id_11":{"attribute_name":"NCID","attribute_value_mlt":[{"subitem_source_identifier":"AN10044054","subitem_source_identifier_type":"NCID"}]},"item_10001_source_id_9":{"attribute_name":"ISSN","attribute_value_mlt":[{"subitem_source_identifier":"0912-0025","subitem_source_identifier_type":"ISSN"}]},"item_10001_text_4":{"attribute_name":"タイトルヨミ","attribute_value_mlt":[{"subitem_text_language":"ja-Kana","subitem_text_value":"ソウセツ
